The digestion behavior of traditional Chinese herbal medicine (TCM) was investigated in orderto enable subsequent determination of potentially toxic elements. Microwave-assisted samplepreparation in high-end quartz vessels in Rotor 8N allows for adequate digestion of a wide range of TCM samples. 1Introduction Traditional Chinese medicine (TCM) is widely used allover the world. The TCM approach is fundamentallydifferent compared to Western medicine. Chinesemedicine has a history of 2000 to 3000 years andincludes therapies like acupuncture, herbal medicineand qigong exercises. One major part of it, the herbal medicine, is based onabout 600 different herbs, which are rarelyadministered singly. Formulations are usually complexmixtures of at least four to twenty different herbs,whereas the exact compositions are often unknown. Harmful concentrations of elements like mercury, leadand cadmium in herbal medicine products are oftendiscussed and sometimes reported. To ensure safedrugs, elemental quantification including a reliabledigestion method is highly requested. 2SSamples The following herbages from the province Shandongwere investigated: Jiao Gu Lan (herb, untreated) Danshen (root,untreated) Licorice (root, untreated) Ginseng(root, untreated) · Cinnamon (cortex, untreated) Ginger (root, untreated) The pre-dried samples were pulverized, using adomestic grinder (Tefal, La Moulinette), packed air-tight and stored in a dry and cool area. Instrumentation The development of the microwave-assisted methodwas carried out in Multiwave 3000, a predecessor ofthe present model Multiwave 5000. Using the same rotors, vessels and accessories themethod is reliably adaptable for Multiwave 5000. During the optimization different sample amounts,reagent mixtures and digestion programs were tested. In the finalized method sample amounts of 500 mgwere digested with 6 mL HNO3. Figure 1: Multiwave 5000 4.1 Digesting the Samples The samples were digested in triplicates, one blankwas added in every microwave digestion run. Thedetected concentrations were mathematicallycorrected for the blank values. Power[Watt] Ramp[min] Hold[min] Fan 1 850 10:00 30:00 1 2 0 0:00 25:00 3 Table 1: Digestion program for 8 vessels Prior to measurement of the digested samples, thesolutions were diluted with deionized water to a finalvolume of either 25 mL or 50 mL. 5 Results 500 mg of TCM herbs or samples with similarmatrices can already be completely digested with 6 mL HNOs and a hold time of 10 minutes only. Apart from the silicate residues, which had noinfluence on the subsequent quantification and weretherefore neglected, clear digestion solutions wereobtained. Except for one sample, the element concentrations inthe seven investigated herbs (Table2) were lowerthan the maximum allowed quantities described in theEuropean Pharmacopoeia. The developed microwave methodprovides the possibility of a quick sample preparationfor subsequent toxicological investigations. Even highsample amounts of 1 g can be completely digestedduring a hold time of only 20 minutes. Seven herbswere measured regarding their cadmium, lead andmercury concentrations, one out of which exceededthe maximum values defined in the EuropeanPharmacopoeia. 7 Reference The method development is the outcome of a projectat the HBLVA 17, Rosensteingasse 79, A-1170 Wienin 2009/2010, under supervision of Prof. JuliusDolischka.
