A Fluorolog® spectrofluorometer, equipped with a 450-W xenon lamp, single-grating monochromators, and near-IR accessories (DSS-IGA020L InGaAs detector and emission grating at 600 grooves mm , blaze = 1000 nm) was used to measure the 1O2 emission spectrum. A high-pass filter (λ = 780 nm) acted as an order-sorter, necessary when such large wavelength-ranges are spanned. Fig. 1 shows the emission spectrum with an integration time of 0.1 s and a bandpass of 12 nm. Note the excellent signal-to-noise ratio.

Singlet-Oxygen Measurements using the Fluorolog@System Introduction The chemistry, spectroscopy, andreactivity of singlet oxygen, 102, is ofconsiderable interest in environmentalapplications, novel organic synthesesand mechanisms of photo-oxygenation.Recently, research has been directedtowardss understanding tthe biologicaleffects of 102, such as thephotodynamic destruction of cells in thepresence of a dye, oxygen, and light. An understanding of themechanisms that produce '02 is neededfor the synthesis of new anti-canceragents used in photodynamic therapy.Spectroscopic properties of the dye and'02mav be characterizedusingfluorescence spectroscopy. The near-IRemission of 102 is around 1280 nm. TheFluorologe system easily detects thissignal with appropriate emissiongratings iand ssiuitable detector.Thestandard R928P photomultipler tuberesponds to photons up to 800 nm.InGaAs detectors extend this range tonearly 1700 nm. The DSS-IGA020L InGaAsdetector is an excellent choice for 'O,measurements. It operates at liquid-N2temperature to minimize thermal noise.Usable from 800-1700 nm, the InGaAsdetector provides a smooth overlap withthe R928P photomultiplier tube. Experimental method and results To demonstrate1'O2 detectiongenerated using a transition-metalcomplex, tris-2'-2'-bipyridineiiruthen-ium(II) chloride, [Ru(bpy)3]Cl2, in D2O, ( ' Wasserman, H . H.; M u rray, R . W., in S i nglet Oxygen; Academic Press: New York, 1979. ) Wasused as a 02 generator. Theoptical density of the solution was 0.3 Aover the D2O baseline at 450 nm. Toenhance theO2 yield, the solution wassaturated with O2 by bubbling pure O2.2through it for 5 min before measure-ment. The Amax of the [Ru(bpy)3]2* com-plex is near 450 nm.
