广州禾信仪器股份有限公司 基于顶空固相微萃取-全二维气相色谱飞行时间质谱联用的唾液与尿液中挥发性成分分析 一、实验目的 采用全二维反向柱系统分别测试吸烟者与不吸烟者的唾液和尿液,对样品进行定性分析,并从中选出能区别吸烟者与不吸烟者代谢物的标志化合物。 二、实验仪器及样品 a)仪器: 设备1:全二维气相色谱-飞行时间质谱联用仪(禾信GGT0610) 设备2:全自动多功能在线前处理进样平台(CTC PAL RTC) b)样品1: C7-C30 正构烷烃混标(购自西格玛奥德里奇,浓度为10ug/mL,溶剂为正己烷); c)样品2:2:H唾液样12个(吸烟者6个样品,编号为 A1- A6;不吸烟者6个样品,编号号B1-B6); d)样品3:3:尿液样12个(吸烟者6个样品,编号为C1-C6;不吸烟者6个样品,编号为D1-D6)。 图1实验仪器实物图 三、实验条件 3.1仪器条件 3.1.1唾液实验条件 a) 前处理进样平台条件: 测试正构烷烃 C7-C30标准品时直接进行液体进样,进样量为 1uL;测试尿液时采用 120um DVB/CWR/PDMS萃取头(用于挥发性和半挥发性物质分析),用移液枪取 3mL 唾液于20mL 固相微萃取样品瓶中。萃取前老化温度为260℃,老化时间为15min, 孵化时间为 30min, 孵化温度为60℃,振摇速度为 350rpm,萃取深度为 22mm, 萃取时间为 50min,进样深度为35mm,解析时间为180s, 解析温度为250℃。 b)色谱条件: 采用前进样口,温度为250℃,分流进样,分流比为2:1;全二维第一根色谱 柱为 MEGA-WAX Plus (30mx0.25mm×0.25um),第二根色谱柱为 DB-17(1.3mx0.18mmx0.18um),载气为氦气,色谱柱恒定流量为 1mL/min; 柱箱采用程序升温,起始温度为40℃,保持3min,,以5℃/min 升温到240℃,保持5min, 共48 min。 c)全二维气相调制器条件: 采用 HV模式,调制柱为 HV调制柱(1.2mx0.25mm),调制周期为5s, 其中解析时间为 1s; N2 补偿气为0.15MPa, 同步 GC 升温程序,调制器进口比柱箱高30℃,调制器出口比柱箱高120℃。 d) 飞行时间质谱条件: 接口温度为320℃,离子源温度为230℃,灯丝2发射电流为200uA,,电离能为标准70eV, MCP 电压为-1920V,采集质量范围为40-500amu,阈值设为90,采集速度为100谱/秒。采集正构烷烃C7-C30标准品溶剂延时为2min,采用 SPME 测试香精样品时不设溶剂延迟。 3.1.2尿液实验条件 a) 前处理进样平台条件: 用移液枪取 10mL 尿液于 20mL 固相微萃取样品瓶中。萃取前老化温度为260℃,老化时间为15min, 孵化时间为30min, 孵化温度为60℃,振摇速度为 350rpm, 萃取深度为 22mm,萃取时间为 50min, 进样深度为35mm,解析时间为180s,解析温度为250℃。 b)色谱条件: 分流比为3:1;其它同唾液条件。 c))全全二维气相调制器条件: 同唾液条件。 d)飞行时间质谱条件: 同唾液条件。 四、数据处理方法 1.采用全二维数据处理工作站软件 Canvas 载入数据,自动绘制全二维 TIC 轮廓图,并对图中信噪比大于3的峰自动识别,标识出的每一个峰点即代表一种化合物,每个化合物由一对保留时间确定,X 轴方向为第一维保留时间(min), Y轴方向为第二维保留时间(s)。 2. 通过对每个化合物的质谱图进行谱库比对检索、结合相对保留指数等信息,对化合物进行定性分析;同时,软件采用峰面积归一化法自动生成各成分的相对含量。根据各化合物的定性定量信息,统计游泥样品中的挥发性成分,并根据化合物化学组成对各个成分进行分类并统计个数和相对百分比含量。 3. 导出定性列表,统计百分比含量较高的组分;统计吸烟者与不吸烟者的唾液与尿液中相对应和不对应的组分。 4. 在吸烟者与不吸烟者的唾液组分对比结果中,将吸烟者与不吸烟者间不同的组分再与各自对照组的组分进行比对,如:将吸烟者组的差异组分与不吸烟者组的所有组分进行比对,若不吸烟者组分中没有与之对应的组分,则说明该化合物为吸烟者特有的标志化合物,反之则不是。 5. 吸烟者与不吸烟者的尿液组分中的标志性化合物筛选方法同唾液筛选方法。 五、结果分析 1.正构烷烃 C7-C30 混标 维二维色谱图,进行峰检测,然后建立RI族,输入对应的保留指数,存为共享模板用于唾液和尿液的辅助定性。全二维数据处理工作站 Canvas 软件可以根据 RI 族自动去计算峰列表里面的所有峰的保留指数,在进行 NIST 谱库检索时,反向柱系统选择标准极性柱,将保留指数误差范围设为30,即可将保留指数用于辅助定性。 2.唾液 2.1吸烟者唾液 吸烟者A1 唾液全二维气相色谱 TIC 轮廓图如图5-2所示,最终共计检测到64种组分。其含量百分比前二十的组分从大到小依次为:吲哚、氨基甲酸苯酯、、二甲基三硫化物、二甲基二硫化物、-1-辛烯-3-醇、1-十二烷醇、对甲酚、 (S)-1-甲基-4-(1-甲基乙烯基)-环己烯、
