This Application Note describes both HPLC and UHPLC methods to simultaneously separate and identify seven tannin compounds as aging markers in barrel-aged alcoholic beverages. The proposed methodology was applied to four different matrixes: whisky, brandy, rum, and tequila, to demonstrate a correlation between aging time and total tannin concentration level. Such a methodology could easily lead to an authenticity verifi cation method to be implemented at the manufacturing process as a quality control measurement, preventing frauds or adulteration.

This Application Note describes a UHPLC/Q - TOF/MS method for the detection of 240 pesticides and pesticide metabolites in diffi cult food matrixes using the Agilent All Ions MS/MS technique. The method benefi ts from increased chromatographic resolution using the Agilent 1290 Infi nity UHPLC system and improved ionization capa bilities with Agilent Jet Stream ionization source. More importantly, the successful screening and quantitation are accomplished by the innate sensitivity improvement of the Agilent 6545 Q - TOF LC/MS system and ion transmission tuning to facilitate the o ptimal ion transmission of small fragile organic molecules. The All Ions MS/MS technique features the easy acquisition method setup and retrospective data interrogation with expanded library. Black tea matrix was chosen for its complexity, and avocado m atrix was chosen to represent food commodities with high lipid content. Our results demonstrate that the improved detection of small organic molecules by the 6545 Q - TOF LC/MS system allows the screening and quantitation of most of the targeted pesticid es below the maximum residue limits (MRLs) specifi ed by regulations

The Agilent 1290 Infinity II LC was used to determine the bitter compounds iso-alpha-acids and reduced iso-alpha-acids, and the clove-like phenolic flavor 4-vinylguaiacol in bottled beer. A six-minute UHPLC method was developed to measure these important solutes in a single run. Diode array and fluorescence detectors (DAD and FLD) were placed in-series for quantification of iso-alpha-acids and reduced iso-alpha-acids, and of 4-vinylguaiacol, respectively. The fluorescence detector provided a selective, sensitive, and accurate determination of ppb amounts of 4-vinylguaiacol. Some figures of merit are presented together with the profiles of a series of different beers.

The Agilent 1290 Infi nity II LC was used to analyze additives such as sweeteners, preservatives, and caffeine in various food products, beverages, and consumer toothpaste. The developed method facilitates accurate and sensitive determination of nine additives using a diode array detector (DAD) and an evaporative light scattering detector (ELSD) placed in series. After the DAD, a valve was installed to remove nonretained solutes in reversed-phase LC, which could increase the noise level of the ELSD. Total analysis time, including re-equilibration, was below 10 minutes with minimal sample preparation. Method performance was evaluated with standard solutions as well as with a series of real samples.

The determination of pesticide residues in food products is important especially for infants and children, as the quantity of food they ingest per kilogram of body weight is relatively high. Some pesticides are known or suspected endocrine disrupting chemicals (EDCs), so the potential for interference with the function of the naturally occurring hormones can severely impact a growing child, even at low concentrations. The implications of long-term, low-dose exposure through food consumption have led to interest in development of highly sensitive analytical methods that combine rapid sample preparation with hyphenated separation and detection. Most pesticide residue laboratories use some variation of the QuEChERS (Quick, Easy, Cheap, Effective, Rugged and Safe) extraction methods. Often the extracted material is analyzed using gas chromatography-tandem mass spectrometry (GC/MS/MS) for the thermally stable, less polar pesticides, or liquid chromatography-tandem mass spectrometry (LC-MS/MS) analysis for the less volatile and/or more polar ones. Concern about the impact on public health from potentially toxic elements and compounds present in everyday foodstuff has led to new legislative guidance. The inorganic forms of arsenic (As) are known to be toxic and carcinogenic to humans, and food and drink are a potential source of exposure [1]. To reduce the intake of inorganic arsenic from the diet, the U.S. Food and Drug Administration (FDA) proposed a new action level for inorganic arsenic in apple juice of 10 μg/L in 2013 [2,3]. Also, following the eighth meeting of the CODEX Committee on Contaminants in Food, the World Health Organization (WHO) proposed a guideline for inorganic As in polished rice of 0.2 mg/kg [4]. There is a clear need for reliable screening methods to accurately determine the levels of such contaminants in food and drink products. The detection limits required are low, as a 0.2 mg/kg limit equates to a digestate concentration of 2 μg/L (assuming a 100x digestion dilution). Even small errors in the precision and accuracy of the analytical method could result in a food product being wrongly classified as safe. Selenium (Se) is an essential micronutrient that can be deficient in the diet as Se-poor soils yield Se-poor food crops. Accurate quantification of Se in food is necessary to assess nutrient status.

A fast and sensitive method using hydride generation (HG) with ICP-MS has been developed for the separation and detection of inorganic arsenic (iAs) in foodstuffs. The iAs measured values in 31 commercial rice products all fell below the current legislative maximum level (ML) of 150 μg/kg set by China. Comparing the results obtained by HG-ICP-MS and HPLC-ICP-MS showed comparable limits of detection were achieved. HG-ICP-MS is ideally suited to the large scale screening of food samples, such as rice, as analysis can be performed in a fraction of the time typically required by a conventional HPLC-ICP-MS approach, with no compromise in analytical performance.

Milk and milk-based products are an important source of nutrients in the human diet, especially for infants and children. Milk products are widely consumed in many parts of the world. Recent developments have extended the analytical dynamic range of the Agilent 7900 ICP-MS to more than ten orders of magnitude, allowing routine measurement of major elements (such as Na, K and Ca) in milk, in the same analytical run as the trace elements.

Red wine produced from the Malbec grape is increasing in popularity in the United States. However, the US is a relatively small producer of the wine compared to Argentina, where it is the most extensively planted grape variety in the country. With rising imports into the US from Argentina there are growing concerns relating to the validation of the geographical origin of this wine. Typically ICP - M S is used to distinguish between wines originating from different regions by comparing the relative concentrations of mineral elements, which are characteristic of the soil composition of the region of production. In this study, a cost - effective approac h has been taken using Microwave Plasma - Atomic Emission Spectroscopy (MP - AES) to measure 6 elements (Sr, Rb, Ca, K, Na and Mg). Agilent’s Mass Profiler Professional (MPP) integrated chemometrics software and another data analysis package were used to m odel the MP - AES results to distinguish the geographical origin of 41 Malbec wine samples produced in Argentina and the USA.

Cereal-based and sucrose-coated infant cereals were ground, and the sucrose, glucose, and fructose concentrations were determined with the Agilent Cary 630 Fourier transform mid-infrared, FTIR, instrument and compared to values measured by HPLC. The degree of substitution for sulfobutyl ether b-cyclodextrin (SBE-b-CD) has historically been characterized by nuclear magnetic resonance spectroscopy, but this approach provides gross values for the degree of substitution. The capillary electrophoresis method described here resolves the mixture of positional and regional isomers based on the degree of sulfo butyl ether substitution. Indirect UV detection has been used, where the detection of cyclodextrin is accomplished by a decrease in background absorbance of the background electrolyte buffer benzoic acid. Ten peaks of SBE-b-CD were well resolved with an optimum pH of 8.0 using an Agilent 50 μm id, 56 cm length bare fused silica capillary. The system suitability requirements were passed as per United States Pharmacopeia (USP), and acceptence criteria for averge degree of substitution 6.2 to 6.9 were met.
