『别名』KLHL14 is a member of the KLHL family. The kelch motif was initially discovered in Kelch. In this protein there are six copies of the motif. It has been shown for one member that it is related to Galactose Oxidase for which a structure has been solved. Two isoforms of this protein exist - 70kDa and 43kDa.

?Kelch样蛋白14抗体洗脱问题

抗体组目前采用的是低pH洗脱方法，按道理说，这个方法应该可以洗脱掉纯大部分抗体，但是有时候出现了一些顽固抗体很难洗脱，大家可以考虑其它的洗脱方法，beads说明书上提到了五大类办法，我觉得比较可靠的方法只有两类，一是改变pH，二是改变离子浓度，另外几种办法要么会带入有机试剂，要么会带入氨基，所以不太适用。改变pH的办法除了用低pH，还可以考虑用高pH来洗脱，好像没多少人尝试过这种办法，在确实没有办法的时候推荐尝试一下。 

?Kelch样蛋白14抗体其它问题

1、看了很多抗体纯化的资料，不管是从血清中纯化多抗还是从腹水中纯化单抗，都推荐大家在上柱子之前把血清或腹水稀释2-5倍，另外把血清或腹水采用0.22微米或0.45微米孔径的膜过滤一次。过滤这一步我们是没条件做的了，但是稀释这一步推荐大家试一试，有文献说稀释可以提高回收率

2、封闭这一步做完后，是不是考虑可以用HCl洗一次？连在beads上的蛋白有的是蛋白间非特异性结合上去的，洗脱之前的任何步骤也许都无法洗干净它们，所以这里把它们都洗掉，以免最后都洗到抗体里去了。

3、CNBr活化的琼脂糖或葡聚糖本身的缺陷：这个非特异性结合影响最大的是和血清中的物质结合了。而血清中大部分蛋白都是免疫球蛋白，其中包括我们要的抗体，它们都带有一定的负电，所以都会和这个部位进行非特异性结合，在洗脱前，有一步是用pH5.0的Gly去洗掉与抗原非特异性结合的抗体的，但是它也会洗掉直接与beads非特异结合的蛋白，也包括我们要的抗体，至于这一步会丢掉多少抗体，好像没有人测过。
