用荧光素标记的试剂进行间接检测的实验步骤(1)将盖玻片、载玻片或培养板置于操作台面。如为1h或1h以下的短程孵育，则勿需湿孵，盖玻片或载玻片可直接放置在实验台上：多个盖玻片则需置于Parafilm膜上以防止抗体溢出盖玻片边缘，这是因为Parafilm孝弹性好，操作时便于用镊子取放盖玻片。但如果圆形盖玻片数量过多，最好将其置于自咆生长和固定的24孔培养板中。如果孵育时间超过1h，则需将其置于培养皿或湿盒中。有数种方法排列盖玻片或戋玻片，以便处理和湿孵，可依具体情况选用。例如，在培养皿上铺一层水饱和的滤纸，再将载玻片置于滤纸上，盖玻片亦可经类似处理。如用24孔培养板，则需在板盖上铺一层潮湿的滤纸来保持湿度。如孵育时间超过1h，可将抗体溶液加在Parafilm膜上的玻片(样品面朝上，再将盖玻片或载玻下)盖在抗体溶液上。孵育完毕后可用PBS洗涤。(2)加入足够量的一抗使其覆盖细胞，但不要溢出载玻片或盖玻片边缘通常加入l0ul。所有稀释液必须用蛋白质溶液配置，如3％BSA／PBS。单克隆抗体通常为组织培养上清液(特异性抗体浓度为20～50txg／ml，未稀释)。腹水、纯化的单克隆抗体和多克隆抗体以及粗制的多克隆血清应该进行稀释度测定。如果特异性抗体的浓度是未知的，测试初始样品的不同稀释度(1：10，1：100，1：1 000，1：10 000)。(3)盖玻片、载玻片或平皿的孵育，在室温下至少30min。对于某些抗原抗体反应，延长孵育时间至12h可提高敏感性。(4)PBS洗3次，5min以上。PBS缓冲液中可以含1％TritonX-100或NP-40，以帮助解决背景问题：(5)加入荧光染料标记的抗Ig抗体。这些试剂应购于商业试剂公司。确认所选择的二抗是特异性针对一抗的种属来源。例如，如果一抗来源于兔，则羊抗兔Ig将是一个很好的选择。二抗应该用含蛋白质的溶液(如3％BSA／PBS)进行稀释。供应商会建议适合的稀释度，但如果没有相关建议，可进行二抗稀释度测定，稀释度范围在1：10和1：1000之间。加入稀释液的竞争蛋白必须为非特异性抗体，其来源应与标记二抗一致，即同一种属的动物。例如，如果使用标记的羊抗兔18，则稀释液中须含1％的非免疫羊Ig，从非免疫动物中分离及纯化。(6)加入带标记的二级试剂后在室温下孵育至少20min，但不应超过1h。(7)用PBS洗3次，每次5min以上。(8)将样品置含有荧光猝灭剂如Gelvatol或Mowiol的甘油溶液中浸泡，在荧光显微镜或共聚焦显微镜下观察并照相。   荧光标记中的注意事项① 为减少非特异性干扰，荧光标记物用前应用0．22um滤膜过滤或高速离心5分钟，以除去颗粒或沉渣。② 细胞样品的制备、染色及保存均应该尽量保持新鲜，防止分解代谢引起的表面抗原消失及细胞死亡，可以通过加入营养培养基或低浓度血清以及4~0或冰浴中操作来解决。③ 细胞稀少或低比例细胞亚群分析时，为保证准确，应排除死细胞。④ 荧光标记抗体浓度应该合适，如果浓度过高，会使非特异性结合增加。在使用一抗之前，将样品与过量的牛血清白蛋白(BSA)或来源于同一种属的正常血清一起孵育，以阻断一抗和细胞表面或胞内结构非特异性作用。在使用一抗之后，将样品、与二抗相同种属的5%～10%正常血清和二抗一起孵育，以减少二抗与一抗、细胞表面或胞内结构之间的非特异相互作用。如果使用含有与一抗或二抗种属相同的血清液体稀释抗体，便可以省略此步骤。
