JASCO日本分光J-1500的MCD磁圆二色应用, ORD and CD can be used to observe these magnetic field-induced optically active molecules. These techniques are more commonly known as magnetic optical rotary dispersion (MORD) and magnetic circular dichroism (MCD). MCD is widely used today to probe the local environment of protein molecules. Chromophores with large magnetic moments arising from either rotational symmetries (aromatics, porphyrins), unpaired spins (metal complexes), or both (hemes) are sensitive to electronic perturbations and therefore provide information regarding the molecule’s electronic state. The MCD signal intensity is proportional to the magnetic field strength, which can be applied by using a permanent magnet.

Biomedicine is a medicinal response to a physical or biological issue with the human body. Biomedicines use active ingredients derived from proteins and the research and development behind these methods is increasing daily. However, these biomedicines are more sensitive to environmental changes, such as a change in temperature, pH, and salt concentration, than traditional pharmaceuticals. To meet the demand for increased sample throughput in the modern pharmaceutical laboratory, JASCO has developed a fully automated high-throughput CD spectral measurement system. This system is composed of a J-1500 CD spectrometer with an autosampler unit. The high-throughput CD system enables the automation of sample pretreatment, injection and washing. In this application note, we will demonstrate the use of the ASU-800 automated system to evaluate the pH dependency on the structure of human serum albumin (HSA)

This application notes demonstrates the use of the J-1500 CD spectrometer and ATS-530 automatic titrator to monitor the interaction of HSA titrated with 3,5-diiodosalicylic acid. While 3,5-diiodosalicylic acid is achiral, its interaction with chiral HSA induces circular dichroism.
