呼吸系统疾病动物模型主要包括慢性支气管肺炎模型，肺气肿模型，肺水模肿型，肺部纤维化、支气管痉挛、哮喘模型，实验性矽肺模型，COPD和急性肺损伤等。

如何评价模型是否成功，常见的方法有：行为学观察，例如哮喘会出现抓耳挠腮、呼吸短促等哮喘发作症状；病理学HE染色观察肺部炎症浸润；免疫组化检测疾病标志，如α-SMA；肺泡灌洗液中的炎症细胞分类计数及ELISA检测细胞因子，如Th2相关细胞因子（IL-4,IL-13等），病理学Masson染色观察纤维增生情况；病理学PAS染色观察杯状细胞增生。

呼吸道内给药是呼吸系统疾病基础研究的关键步骤，由于不同动物类型，不同造模条件的反应不同，造模方式各异，给药方法亦是多种。以往有环甲膜穿刺法，气管切开法等。环甲膜穿刺给药动物死亡率低，但定位困难，加之给药部位偏上，给药后动物呛咳常常使得药液回流至上呼吸道，而不能保证肺部的药量，影响结果。气管切开给药虽然可以保证药量及药物在肺部的均匀分布，但手术创伤大，死亡率高，不是理想的给药方法。

上海交通大学医学院附属瑞金医院&荷兰格罗宁根大学医学中心科研团队在著名期刊Advanced Science（2022影响因子17.521、SCI 1区）DOI:10.1002/advs.202200856 发表《Autologous Skin Fibroblast-Based PLGA Nanoparticles for Treating Multiorgan Fibrosis》。

在本研究中，8周大的C57BL/6小鼠在实验之前用异氟醚麻醉，肺纤维化是由博来霉素使用北京元森凯德生物技术有限公司（YSKD biotechnology,Beijing）研制生产的动物雾化吸入相关仪器递送到小鼠肺部，小鼠每5天给药FNPs，RNPs, 并通过三个具有各种器官纤维化的独立动物模型确认了FNPs 体内的抗纤维化功效，表明其重要的临床潜力。

通过超声波图像的综合强度来量化肝纤维化严重程度（n = 每组6只生物独立的小鼠）。来自不同治疗组的小鼠的血清肝功能测试（n = 每组6只生物独立的小鼠）。促纤维化、促炎、ECM交联相关和肝脏代谢相关基因表达的如上图所示。

肺纤维化（ pulmonary fibrosis，PF）是一种间质性肺疾病，以巨噬细胞、淋巴细胞等炎症细胞在肺间质浸润、纤维母细胞增生及纤维结缔组织沉积于肺间质致使出现弥散性肺泡炎和肺泡结构紊乱，并最终导致肺间质纤维化为特征，是一系列慢性肺部疾病的最终结局。因为肺纤维化的体外研究受到条件的限制，目前基本没有合适的研究方法，因此建立与人肺纤维化病理过程相似的动物模型尤为重要。
