B/c-ES细胞；小鼠ES细胞

B/c-ES细胞；小鼠ES细胞实验方法如下：

1、从手术室无菌取脑髓灰质或白质后，仔细剥除脑膜、血管和纤维成分，置Hanks液中漂洗一、二次。

2、置于30—50倍体积的Hanks液中，此时脑组织比较柔软，反复吹打即制成细胞悬液。

3、把悬液注入离心管室温中直立5—10分钟后，细胞或细胞团快自然下沉，脂肪等杂物易漂浮，可吸除上层、反复二、三次，即可排除脂肪成分和其它碎块并获较多细胞成分。

4、在沉降物中加入适量培养液，通过纱网成纱布过滤，计数并调整细胞密度。

5、接入培养瓶或皿中，置5%CO2温箱中培养。

B/c-ES细胞；小鼠ES细胞培养准备和安装过滤器：清洗好过滤器，干燥，放入一张孔径0.22μm的微孔滤膜，用布包装好，15磅20 min进行高压灭菌处理。 在超净台内打开过滤器架好，胶管一端接入滤泵再插入待除菌的液体中，出口端胶管深入到已消毒好的瓶子中。用泵过滤，过滤后要检查滤膜是否完好无损。

B/c-ES细胞；小鼠ES细胞在适宜培养基中可以增殖，从而可以测定肿细胞克隆形成率。造血系统软琼脂集落形成试验方法相同，主要用于有关细胞分化的研究，但使用培养基不同。
