    [B][center]flash色谱简介[/center][/B]1978年，Sill发明了FLASH色谱，文章发表在J.O.C上，从此FLASH色谱方法得以建立，新的设备也不断涌现，在国内，大连理工大学的彭勤纪教授自国外归国后，引进了该技术并进行了系统综述，发表在《化工进展》上，该文章首次将FLASH色谱译为“快速色谱”。狭义的快速色谱是指用压缩气体产生压力，加速流动相洗脱速度的低压短柱制备液相色谱系统，广义的快速色谱系统是指一切输液方式的中低压液相色谱系统。快速色谱有人还译为“闪式色谱”“FLASH色谱”。经过20余年的发展，FLASH色谱已经广泛被人使用，成为常规的纯化分离设备，如葛兰素公司，辉瑞公司的研究人员都拥有数百套FLASH色谱装置，世界上也有多家仪器厂商提供全套的FLASH色谱装置。FLASH色谱脱胎于开口玻璃柱色谱，主要用于天然产物，有机合成产物的分离纯化。古典的FLASH色谱由玻璃柱，贮液瓶，压缩空气调节阀组成，Aldricn公司，Baker公司都有相应的产品。该类型成本低，便于观察，但压力低，易破损。现在许多实验室在玻璃柱子顶端放置一个加压皮球，属于同类产品。经过改进，形成了经典FLASH色谱系统。由输液压力罐、流量调节阀、FLASH预装柱、上样装置组成，设备紧凑，压力高，使用方便，重现性高。经典的FLASH色谱仍使用气压加压，压力范围约0-7bar，柱长约15cm，填料粒度为40～60。可固体与液体上样，手工收集流分。 最近，因为科研竞争的加剧与实验室人力成本的增长，要求更高的自动化程度与分离速度，产生了自动化FLASH色谱，装置带有柱、色谱泵、检测器、自动馏分收集器、工作站，有的仪器还有梯度洗脱系统与柱切换系统、自动进样系统，可自选优化条件，实现无人操作，极大的降低了人力成本，加快了开发速度。目前国内有人使用蠕动泵输液，用有机玻璃柱分离，严格上说不能于用有机相小分子分离，只能用于生化分离，否则会损坏仪器。色谱用于制备，拥有以下优点：1、速度快，一般为开口柱色谱分离时间的1/8，节省时间，样品变化分解慢2、制备量大，比同样直径的HPLC制备柱，至少可多上一倍量样品。3、成本低，首先仪器设备低于HPLC；其次，使用溶剂比玻璃柱节省，硅胶柱可多次使用，节省硅胶用量。4、使用简单，不需要特殊溶剂处理，样品可以干法上样，人员不需要经过特殊培训。色谱的效果主要取决于洗脱溶剂的优化与柱子装填效果，在处理与制备HPLC关系时，要遵循以下原则：（1）能只用FLASH色谱时，就不用制备HPLC（2）要先用FLASH色谱，纯化再用制备HPLC其它的使用技巧还有：①先用TLC选反溶剂、负载量②使用短粗柱(直径：长度≤1：7)[B]flash色谱柱：[/B][img]http://ng1.17img.cn/bbsfiles/images/2009/09/200909142001_171059_1600062_3.jpg[/img]特点：1.可用相位：C8, C18, Silica, NH2, CN, Alumina Acidic, Alumina Neutral, Alumina2.尺寸：Biotage兼容12S, 12M, 40S, 40M, 40L, 65i, 75L, 75M, 75L, 150M and 150L3.能够承受100ps的压强[B]flash色谱图：[/B][img]http://ng1.17img.cn/bbsfiles/images/2009/09/200909142003_171060_1600062_3.gif[/img]
