Enzymic procedures vary in pre-treatment steps,2 starchgelatinisation, liquefaction and dextrinisation, hydrolysis of dextrins toGlu cose and Glu cose measurement3. AACC Method 76-114 specifiesstarch gelatinisation under aqueous conditions in an autoclave,followed by starch conversion to Glu cose with amyloglucosidase andGlu cose measurement. AACC Method 76-11 underestimates starchcontent in a range of samples and materials, including high amylosemaize starches and many processed cereal products. Most methods inuse today incorporate treatment with thermostable α-amylase eitherduring or immediately following the starch gelatinisation step.5,6 Forsamples which are difficult to gelatinise (such as high amylose maizestarch) solvents such as sodium hydroxide or dimethyl sulphoxide(DMSO)7,8 have been employed. In a procedure to ensure completesolubilisation of starch in dietary fibre determination, Englyst andCummings (1988)7 included treatment with the starch debranchingenzyme, pullulanase.To satisfy the need for an extremely simple, yet quantitative andreliable, procedure for the measurement of total starch Megazymeproduces, and offers, a total starch assay kit based on the use ofthermostable α-amylase and amyloglucosidase (McCleary et al.9). Thismethod has been adopted by AOAC (Official Method 996.11) andAACC (Method 76.13).More recently, thermostable α-amylases that are active and stableat lower pH values have become available. Consequently, we haveupdated our total starch methodology to incorporate such anenzyme10. The major advantage of this improvement is to allow boththe thermostable α-amylase and amyloglucosidase incubation stepsto be performed at the same pH (pH 5.0) which, in turn, simplifiesthe assay and minimises the possibility of production of maltulose(4-α-glucopyranosyl-D-fructose)11, which is resistant to hydrolysis byamyloglucosidase and α-amylase.The Megazyme total starch analysis procedure allows themeasurement of total starch in a wide range of food, feed, plant andcereal products (natural or processed). For most samples (e.g. wheatflour), starch is completely solubilized on incubating the sample atapprox. 100°C in the presence of thermostable α-amylase. PH值标准缓冲液:PH值为 4.0,7.0 和10.0。6. 酶的纯化和标准化6.1 根据AACC/AOAC的膳食纤维检测方法,必须确保酶的纯度。6.1.1 淀粉葡萄糖苷酶被木聚糖酶、纤维素酶和果胶酶污染,会低估β-葡聚糖、阿拉伯木聚糖和果胶的含量。6.1.2 热稳定a-淀FEN酶被污染,会影响抗性淀粉的测量。6.1.3 蛋白酶被4-β-葡聚糖酶污染,会低估β-葡聚糖的含量。6.1.4 U/mL,1个酶活力单位定义为:40°C,PH值4.5时,每分钟释放1 μmol葡萄糖所需要的酶量。--- 酶活力表示方法2:对-硝基苯基-b-麦芽糖苷(PNPBM)法,200 U/mL, 1个酶活力单位(1PNP单位))定义为:40°C,有过量萄糖苷酶存在下,每分钟从对硝基苯基-β-麦芽糖苷释放1 μmol 对硝基苯基所需要的酶量。6.2.2 热稳定a-淀FEN酶,0.05 mL,--- 酶活力表示方法1:以淀粉为底物,以Nelson/Somogyi还原糖表示,10000U/mL ,1个酶活力单位定义为:40°C,PH值6.5时,每分钟释放1 μmol葡萄糖所需要的酶量。--- 酶活力表示方法2:对硝基苯基麦芽糖为底物,3,000 U/mL(Ceralpha),1个酶活力单位定义为:40°C,PH值6.5时,每分钟释放1 μmol对-硝基苯基所需要的酶量。6.2.3 蛋白酶,0.10 mL--- 酶活力表示方法1:酪蛋白测试,350 U/mL或50 mg/mL或7 Tyrosine U/mL,1个酶活力单位定义为:40°C,PH值8.0时,每分钟从可溶性酪蛋白中水解出(并溶于三氯YI酸)1 μmol酪氨酸所需要的酶量。--- 酶活力表示方法2:偶氮酪蛋白测试,350 U/mL或50 mg/mL或7 Tyrosine U/mL,1个内肽酶活力单位定义为:40°C,PH值8.0时,每分钟从可溶性酪蛋白中水解出(并溶于三氯YI酸)1μmol酪氨酸所需要的酶量。该偶氮酪蛋白(货号:S-AZCAS).7. 检测步骤7.1 试样制备准确称取双份试样各1 g,质量差小于0.005g,精确到0.1mg,置于400mL或600mL高型烧杯中,同时制备双份空白样,在每个烧杯中加入40 mL PH值8.2的MES-TRIS缓冲液,磁力搅拌,直到试样完全分散到缓冲液中。7.2 热稳定a-淀FEN酶酶解:加入50 μL热稳定a-淀FEN酶溶液,加盖铝箔,置于95-100°C恒温振荡水浴中持续振摇,当所有烧杯全部放入水浴开始计时,反应35分钟。7.3 冷却:将烧杯取出玲却到60°C,除去铝箔盖,用刮勺将烧杯内壁和底部的胶状物刮下,用10 mL蒸馏水冲洗烧杯壁和刮勺。7.4 蛋白酶酶解:在每个烧杯中各加入100 μL蛋白酶溶液,加盖铝箔,置于60°C恒振荡水浴中,当烧杯内温度达到60°C开始计时,持续反应30分钟。7.5 PH值调节:反应30分钟后,边搅拌边加入0.561 mol/L盐酸,严格控制60°C,用 1mol/L氢氧化钠溶液或1mol/L盐酸溶液,控制PH值在4.5±0.2。7.6 淀粉葡萄糖苷酶酶解:在上述溶液中边搅拌边加入200 μL淀粉葡萄糖苷酶溶液,加盖铝箔,置于60°C恒温振荡水浴中,持续振摇,当烧杯内温度达到60°C开始计时,反应30分钟。8. 测定8.1 不溶性膳食纤维测定:8.1.1 过滤洗涤:试样酶解液全部转移到坩埚中过滤,残渣用10mL预热到70°C的蒸馏水洗涤两次,合并过滤液,转移至另一600mL高脚烧杯中,备测可溶性膳食纤维。残渣分别用10mL 95%乙醇和丙酮洗涤两次,抽滤去除洗涤液,将坩埚连同残渣在103°C烘干过夜,将坩埚置于干燥器中冷却1小时,称重(包括坩埚、膳食纤维残渣和硅藻土),精确至0.1mg,减去坩埚和硅藻土的干重,计算残渣质量。8.1.2 蛋白质和灰分测定:称完质量的残渣和硅藻土的混合物,一份用凯氏定氮法,以N×6.25为换算系数,计算蛋白质质量。另一份在525°C灰化5小时,于干燥器中冷却,精确称量坩埚总量(精确至0.1mg)减去坩埚和硅藻土干重,计算灰分质量。8.2 可溶性膳食纤维测定8.2.1 计算滤液体积:将不可溶性膳食纤维过滤后的滤液收集到600mL的高型烧杯中,通过烧杯+滤液总重扣除烧杯质量的方法估算滤液的体积。8.2.2 沉淀:将滤液加入4倍体积预热60°C的95%乙醇,或取80g滤液加入320mL预热60°C的95%乙醇室温下沉淀1小时。8.2.3 过滤:在干燥的坩埚中加入1g硅藻土,70°C真空干燥至恒重,记录坩埚的质量(精确到0.1mg)。用15mL 78%乙醇将硅藻土润湿,并用真空溶剂过滤装置在抽真空条件下,使硅藻土平铺于坩埚中。将样品酶解液缓慢转移至对应的坩埚中,抽滤。用刮勺和78%乙醇将所有残渣转至坩埚中。8.2.4 洗涤:分别用15mL 78%乙醇、15mL 95%乙醇和15mL丙酮洗涤残渣各两次,抽滤去除洗涤液后,将坩埚连同残渣在105°C烘干过夜。将坩埚置于干燥器中冷却1小时,称重(包括坩埚、膳食纤维残渣和硅藻土),精确至0.1mg。减去坩埚和硅藻土的干重,计算残渣质量。8.2.5 氢氧化钠溶液(0.275mol/L):去11g氢氧化钠溶解在700mL蒸馏水中,冷却转移到容量瓶中,用水定容到1L。f. 盐酸溶液(0.325 mol/L):取 325 mL 1.0 mol/L 盐酸置于容量瓶中,用水定容到1L。3. 样品准备总膳食纤维的测定必须是低脂或无脂的干燥样品,取混匀后的样品于70°C真空干燥过夜,干燥器中冷却,再称重记录重量损失,干样粉碎后过0.3-0.5 mm筛,若试样不能加热,则冷冻干燥后再粉碎过筛。如果是高脂肪样品(> 10 %),取经过干燥的样品分别用25mL石油醚脱脂三次,干燥后记录脱脂的质量损失,最后计算总膳食纤维含量时进行进行校正,粉碎过筛后的干燥试样放在干燥器中待用。4. 分析步骤准确称取双份试样各1 g,质量差小于0.005g,精确到0.1mg,置于400mL或600mL高型烧杯中,同时制备双份空白样,在每个烧杯中加入50 mL PH值6.0的磷酸盐缓冲液,磁力搅拌,直到试样完全分散到缓冲液中,控制PH值6.0±0.1。4.2 热稳定a-淀FEN酶酶解:加入50 μL热稳定a-淀FEN酶溶液,加盖铝箔,置于沸腾水浴中15分钟,每5分钟轻轻振荡一次。当烧杯内的温度到100°C开始计时,总共大约30分钟。4.3 冷却:将烧杯取出冷却到室温,加入10 mL 0.275mol/L 的氢氧化钠,调节PH值在7.5±0.1。4.4 蛋白酶酶解:在每个烧杯中各加入100 μL蛋白酶溶液,加盖铝箔,置于60°C恒振荡水浴中,当烧杯温度达到60°C开始计时,持续反应30分钟。4.5 冷却:加入10 mL 0.325 mol/L盐酸,调节PH值在4.5±0.2。4.6 淀粉葡萄糖苷酶酶解:在上述溶液中边搅拌边加入200 μL淀粉葡萄糖苷酶溶液,加盖铝箔,置于60°C恒温振荡水浴中,持续振摇,当烧杯内温度达到60°C开始计时,反应20分钟。4.7 在每份试样中加入预热至60°C的95%乙醇280mL(预热前的体积),乙醇与样液体积比为4:1,取出烧杯,盖上铝箔,室温下沉淀1小时。4.8 过滤:在干燥的坩埚中加入1g硅藻土,70°C真空干燥至恒重,记录坩埚的质量(精确到0.1mg)。用15mL
