Human herpesvirus 5 strain UL32-EGFP-HCMV-TB40 is propagated in MRC-5 cells (ATCC CCL-171). This strain was sourced from the in vitro recombination of HCMV strain TB40 with a plasmid carrying the TB40 UL32 gene fused to GFP. Because GFP is associated with a structural protein, virus particles fluoresce green under appropriate illumination; this is useful for following intracellular traffic of virus particles.

Incubation
7-21 days at 37°C in a humidified 5% CO2 atmosphere, until CPE is progressed through 80% of the monolayer.
Recommendation for infection
Plate cells 24-48 hours prior to infection and infect when cultures are 80-90% confluent. Remove medium and inoculate with a small volume of virus (eg, 1 mL per 25 cm2) diluted to provide an optimal MOI (eg, 0.1 to 1.0). Adsorb 1-2 hours at 37°C in a humidified 5% CO2 atmosphere. End adsorption by adding virus growth medium.
