前列腺癌多组学分析领域的应用

复旦大学姜昊文、党永军等研究团队在癌症诊断治疗方面实现新进展,在学术期刊《Nature:signal transduction and targeted therapy》上发表了题为“Integrative multi-omics and drug–response characterization of patient-derived prostate cancer primary cells” 的文章(影响因子IF=39.3),研究人员称他们的综合多组学方法可以全面了解前列腺癌的生物标志物和药理反应,从而实现更精确的诊断和治疗。

这篇文章研究了来自 35 例中国前列腺癌(PCa)和良性前列腺增生(BPH)患者的原发性细胞模型,建立了前列腺癌细胞库(PCMR)。通过对PCMR进行基因组、转录组、全蛋白组和表面蛋白组分析,揭示了多层次分子特征。通过药物蛋白相互作用分析,发现 AGR2 表达下调可以增强 Crizotinib 的抑制活性,机制可能是通过 ALK/c-MET-AKT 轴。其中有采用Clinx勤翔一体式化学发光成像仪ChemiScope S6进行蛋白印迹成像与分析。



改善药物滥用新策略研究中应用

江苏神经精神疾病重点实验室镇学初和徐林教授团队在《Signal Transduction and Targeted Therapy》上发表了题为“Hypoxia-inducible factor upregulation by roxadustat attenuates drug reward by altering brain iron homoeostasis” 的文章(影响因子IF=39.3)

研究揭示了罗沙司他上调缺氧诱导因子,通过改变脑中铁稳态来减弱药物奖励,这可能是治疗药物滥用的一种潜在的新治疗策略。研究结果还通过提出治疗药物使用障碍的潜在新适应症,重新调整了Rox的用途。其中,研究团队使用Clinx勤翔化学发光成像ChemiScope3300Mini设备进行蛋白印迹成像与分析。



构建免疫缺陷猴模型研究中的应用

暨南大学粤港澳中枢再生研究院闫森和涂著池团队在《Signal Transduction and Targeted Therapy》上发表了题为”Generation of inactivated IL2RG and RAG1 monkeys with severe combined immunodeficiency using base editing“的文章(影响因子IF=39.3)

成功地使用胞嘧啶碱基编辑器(CBE)4max系统构建了一种能够支持肿瘤生长的免疫缺陷猴模型。这些免疫缺陷的猴子是临床前研究的有价值的工具,并弥合了小动物模型和人类之间的差距。利用它们可以显著提高临床前研究在抗癌药物开发和异源细胞或器官移植方面的效果。其中,研究团队使用Clinx勤翔化学发光试剂盒检测蛋白条带。



真核生物线粒体功能研究领域的应用

中国科学技术大学张亮,施蕴渝团队在在学术期刊《Nucleic Acids Research》上发表题为“The recognition mode between hsRBFA and mitoribosome 12S rRNA during mitoribosomal biogenesis”的文章(影响因子IF=14.9)。该项工作系统地研究了人源线粒体核糖体结合因子(hsRBFA)识别线粒体核糖体小亚基12S rRNA的分子机理及其对线粒体核糖体的成熟和线粒体功能的影响。

实验结果显示,hsRBFA通过其N末端和KH结构域与12S rRNA相互作用。通过电泳迁移实验发现,hsRBFA的KH结构域单独存在时无法与12S rRNA的28结构螺旋、44结构螺旋和45结构螺旋以及3'末端区域结合,而将N末端加入KH结构域后,可以与上述区域均有结合。此外,hsRBFA的C末端对RNA识别无效。进一步实验证实,hsRBFA的近全长片段和N末端-KH同样可以与12S rRNA区域结合,而仅含有KH结构域和C末端的片段无法结合任何RNA底物。总之,hsRBFA通过其N末端和KH结构域二者共同作用与12S rRNA相互作用,其中N末端起主要作用。通过对hsRBFA不同结构域与12S rRNA亲和性的检测比较,该小节主要阐明了hsRBFA是通过其N末端和KH结构域共同结合12S rRNA的结论。其中,有使用Clinx勤翔GenoSens 2000凝胶成像系统,显示了 hsRBFA 与 12S rRNA 之间的相互作用,为探究 hsRBFA 的哪个位点主要与 RNA 结合提供依据。



小鼠非酒精性脂肪性肝炎治疗领域的应用

山东第一医科大学秦树存教授和上海交通大学何前军教授团队合作在《Theranostics》上发表题为“A strategy of local hydrogen capture and catalytic hydrogenation for enhanced therapy of chronic liver Diseases”的文章(影响因子IF=12.4)

研究报道了使用Pd纳米颗粒预防和治疗小鼠非酒精性脂肪性肝炎(NASH)的治疗策略。该策略基于Pd纳米颗粒催化加氢的特性,实现了肝脏靶向高剂量递送氢分子,为寻求安全高效的慢性肝病(CLD)氢分子疗法开辟了一条新道路。其中,研究团队使用Clinx勤翔荧光及化学发光成像系统ChemiScope 6200Touch进行蛋白印迹成像与分析。



间充质干细胞加速烧伤创口愈合方面的应用

海军军医大学第一附属医院烧伤科肖仕初课题组在《Materials Today Bio》上发表题为“Acceleration of burn wound healing by micronized amniotic membrane seeded with umbilical cord-derived mesenchymal stem cells”的文章(影响因子IF=8.2)

作者构建了mAM-MSC复合物,并在烧伤-伤口模型中评估了它们的促愈合和血管生成作用。研究发现mAM可以在体外实现有效的MSC扩增,并在体外和烧伤伤口中保持较高的细胞活性。mAM-MSC在烧伤创面上的应用可显著提高烧伤创面的愈合率和血管数量。其中,研究团队使用Clinx勤翔小动物活体成像系统IVScope 8500评估移植的mAM-MSC在小鼠烧伤伤口中的生存时间。



急性心力衰竭治疗领域的应用

海军军医大学梁晓及美国威斯康星大学 Hector H. Valdivia 团队在学术期刊《Materials Today Bio》上发表题为“OpiCa1-PEG-PLGA nanomicelles antagonize acute heart failure induced by the cocktail of epinephrine and caffeine”的文章(影响因子IF=8.2)

这篇文章研发及评价了一款新型 OpiCa1-PEG-PLGA 纳米载体系统。结果表明其具有良好的靶向性、稳定性和低毒性,令人信服地作为新的疾病治疗途径。文章系统阐述了各项研究方法与结果,对提高理解 OpiCa1 的作用机制与纳米药物学理论具有重要参考价值。其中,研究人员使用Clinx勤翔 IVScope 8000小动物活体成像系统跟踪观察了纳米颗粒在体内的分布情况。
