Apoptosis-inducing natural products found in utero during murine pregnancy

A family of polyunsaturated fatty acids is shown to be abundant in utero during pregnancy. Members of this family are able to induce cleavage of poly(ADP)ribose polymerase, and ultimately to induce apoptosis, in T-antigen-transformed Jurkat T cells. Free radical scavengers, including phenol and benzyl alcohol, block the apoptosis-inducing properties of these polyunsaturated fatty acids; this is consistent with a lipid peroxidation mechanism involving formation of hydroperoxy fatty acids.

Enantioselective Synthesis of S-Equol from Dihydrodaidzein by a Newly Isolated Anaerobic Human Intestinal Bacterium

A newly isolated rod-shaped, gram-negative anaerobic bacterium from human feces, named Julong 732, was found to be capable of metabolizing the isoflavone dihydrodaidzein to S-equol under anaerobic conditions. The metabolite, equol, was identified by using electron impact ionization mass spectrometry, 1H and 13C nuclear magnetic resonance spectroscopy, and UV spectral analyses. However, strain Julong 732 was not able to produce equol from daidzein, and tetrahydrodaidzein and dehydroequol, which are most likely intermediates in the anaerobic metabolism of dihydrodaidzein, were not detected in bacterial culture medium containing dihydrodaidzein. Chiral stationary-phase high-performance liquid chromatography eluted only one metabolite, S-equol, which was produced from a bacterial culture containing a racemic mixture of dihydrodaidzein. Strain Julong 732 did not show racemase activity to transform R-equol to S-equol and vice versa. Its full 16S rRNA gene sequence (1,429 bp) had 92.8% similarity to that of Eggerthella hongkongenis HKU10. This is the first report of a single bacterium capable of converting a racemic mixture of dihydrodaidzein to enantiomeric pure S-equol.

New Spirorotenoids from Tephrosia candida

The ethyl acetate extract of roots of Tephrosia candida afforded three new spirorotenoids belonging to a new class of spirocompounds, named tephrospirolactone, tephrospiroketone I, and tephrospiroketone II. The structures of these compounds were determined mainly based on spectral analysis. The only known spirorotenoid described in the literature is amorphispironone, isolated from Amorpha fruticosa.

Electronic Interactions in a New Fullerene Dimer: C122H4, with Two Methylene Bridges

The isolation of a new fullerene dimer, C122H4, and its structural characterization by 13C NMR and 1H NMR spectroscopy and by UV/vis and IR spectroscopy are reported. The structure of this dimer consists of two fullerene cages, which are directly connected through two C-C bonds and two methylene bridges. Consequently, adjacent hexagonal faces of the two fullerene cages are arranged in a face to face manner. Molecular orbital calculations indicate that the proximity of the fullerene cages results in significant through space overlap in both the HOMO and LUMO. As a consequence of this overlap, the electrochemistry of the dimer shows electronic communication with stepwise reduction of each cage.
