In Search of Calorimetric Answers to Protein Folding, Ligand Binding, Aggregation and Molecular Partitioning in Self Assemblies

OVERVIEW
Isothermal titration (ITC) and differential scanning calorimetry (DSC) are powerful tools for thermodynamic characterization of protein folding intermediates, drug-protein interactions, protein-solvent interactions, and prevention of fibrilization of proteins. In combination with other techniques such as light scattering and x-ray crystallography, the examination of the influence of charge, water molecule participation and the complex plaque and fibril formations are revealing many underlying structural features and their functions in diseases where they have been observed. Examples of molecular binding events and the importance of the water molecules to characterize specificity will be shown. In addition, examples using DSC and light scattering to describe structural stability and its influence on the formation of fibrils and plaques will be discussed. Throughout the presentation details of experimental techniques, instrument requirements and in-depth data interpretations show the power and flexibility of microcalorimetry in many areas of biological research.
ABOUT THE SPEAKER
Professor Nand Kishore
Prof. Kishore earned his PhD (Chemistry) from the Indian Institute of Technology Delhi in 1989, followed by postdoctoral training at Yale University, U.S.A. (1989-1991) and at the National Institute of Standards and Technology, U.S.A. (1991-1992). He joined as faculty at IIT Bombay in 1992. His research interests include understanding physical chemistry underlying the biologically important systems. His research group has contributed to the thermodynamic characterization of protein folding intermediates, drug-protein interactions, protein-solvent interactions, and prevention of fibrilization of proteins by using a combination of calorimetric and spectroscopic techniques. He is on the editorial board of The Journal of Chemical Thermodynamics and a Fellow of the National Academy of Sciences, India. He was President of the Indian Chemical society, Mumbai Branch from 2010 to 2012. He has received The Rev. Fr. L. M. Yedanapally Memorial Award for the Year 2011 for contribution to research in Physical chemistry by Indian Chemical Society. He has published about 100 papers in international journals in the areas of Physical Chemistry, Biophysical Chemistry and Biochemistry.
